Ca²⁺ plays a vital role in the blood coagulation process, mainly including the following aspects:
1. Participation in the activation of coagulation factors:
Many coagulation factors require the participation of Ca²⁺ when they play a role. For example, in the process of activation of coagulation factors IX, X, XI, XII, etc., Ca²⁺ is required to bind to these coagulation factors to make them undergo conformational changes and expose the active center, so that they can interact with other coagulation factors and start the coagulation cascade reaction.
2. Promote the formation of coagulation factor complexes:
Ca²⁺ can act as a bridge to promote the formation of complexes between coagulation factors. For example, in the coagulation process, Ca²⁺ can connect negatively charged phospholipids (present on the surface of platelet membranes, etc.) with coagulation factors Xa, V, etc. to form prothrombin complexes, accelerating the process of prothrombin conversion to thrombin.
3. Platelet activation and release:
Ca²⁺ is also very important for the activation and release reactions of platelets. When blood vessels are damaged, platelets adhere to the damaged part, and Ca²⁺ enters the platelets, causing a series of biochemical reactions in the platelets, leading to the release of a variety of bioactive substances from the platelets, such as adenosine diphosphate (ADP), thromboxane A₂, etc. These substances further promote platelet aggregation and blood coagulation.
4. Stabilize fibrin polymers:
In the final stage of coagulation, fibrinogen is converted into fibrin monomers under the action of thrombin, and then the fibrin monomers are cross-linked to form stable fibrin polymers under the action of Ca²⁺ and coagulation factor XIII, thereby forming a solid blood clot to achieve the purpose of hemostasis. Without Ca²⁺, fibrin monomers cannot be cross-linked into stable fibrin polymers, blood clots cannot be effectively formed, and blood cannot coagulate normally.
